Piroxicam is a nonsteroidal anti-inflammatory drug (NSAID) belonging to the oxicam group. It is renowned for its analgesic, anti-inflammatory, and antipyretic effects and is widely used in clinical practice. Piroxicam exerts its therapeutic effects by inhibiting the enzyme cyclooxygenase (COX), which is responsible for synthesizing prostaglandins—key substances that mediate inflammation, pain, and fever.
Chemical Structure and Properties
Piroxicam has the chemical name 4-hydroxy-2-methyl-N-(2-pyridinyl)-2H-pyridine-3-carboxamide. Its molecular formula is C₁₅H₁₃N₃O₃, with a molecular weight of approximately 275.28 g/mol. It appears as a white to off-white crystalline powder, slightly soluble in water but more soluble in organic solvents such as ethanol
Mechanism of Action
Piroxicam works by inhibiting cyclooxygenase (COX), thereby reducing the synthesis of prostaglandins. There are two main isoforms of COX: COX-1 and COX-2. COX-1 is primarily involved in normal physiological functions, such as protecting the gastric mucosa and regulating platelet aggregation, while COX-2 is induced during inflammation and is responsible for producing inflammatory prostaglandins. Although Piroxicam inhibits both COX-1 and COX-2, its selectivity for COX-2 is lower compared to some other NSAIDs (such as Meloxicam). By reducing prostaglandin synthesis, Piroxicam effectively alleviates pain, reduces inflammation, and lowers fever.
